A research group designing a 3rd-generation CAR-T construct targeting CD19. The Design tool walks through scFv selection (VH/VL from FMC63), CD8α hinge, CD28-TM, CD28+4-1BB co-stimulatory domains, and CD3ζ. The assembled construct is codon-optimized for human expression, RE-scanned against pLEV113 MCS, and exported as a complete sequence ready for gene synthesis.
Engineering a TGFβRII–PD-L1 checkpoint-blocking decoy fusion protein. Domain A (TGFβRII ECD) is looked up via UniProt, pasted into Design, connected to Domain B (PD-L1 IgV) via a (G₄S)₃ flexible linker, secretion signal added. CAI, GRAVY score, and predicted pI are calculated. Codon-optimized DNA ready for CHO expression is exported.
Designing a CD3×CD19 BiTE construct. Both VH/VL pairs entered in the Bispecific workspace, BiTE format selected, intra-scFv (G₄S)₃ and inter-scFv (G₄S)₃ linkers configured. Full construct assembled, immunogenicity screened via IEDB link, and structure predicted via ABodyBuilder2. Sequence exported for E. coli periplasmic expression in pET-28a.
After designing and cloning a construct, the Develop tool manages the full experimental campaign: materials ordered, SOP-referenced protocol steps logged, flow cytometry results entered with EC50 values, and a structured technical report generated for sign-off by the PI.
